B1, B2, or both
B1 covers business travel — meetings, negotiations, a conference, a trade fair. B2 covers tourism, family visits and medical treatment. Most applicants are issued the combined B1/B2, but you still have to pick a primary purpose and be consistent about it from the DS-160 through to the interview desk.
Neither category allows you to work in the United States, and answering as though it might is one of the quicker ways to lose the application.
The DS-160 is part of the interview
The DS-160 is a long online form, and applicants treat it as paperwork to be finished. The consular officer has it open on screen while they talk to you. Every inconsistency between what is typed there and what you say out loud is visible to them instantly.

Appointments in Pakistan
Pakistani applicants are seen at the US Embassy in Islamabad or the Consulate General in Karachi. Booking runs through the official appointment service after the MRV fee is paid, and wait times swing a great deal through the year — early booking matters more than almost anything else you can control.
Some renewal applicants qualify to submit without appearing in person. Whether you do depends on your previous visa and how recently it expired, and it is worth checking before you assume you need an interview slot at all. If you are travelling for urgent medical or humanitarian reasons there is a separate expedited request route, which is granted on evidence, not on urgency alone.
